Here we demonstrate that cysteine sulfinic acid labeling with the alkyne-functionalized electrophilic diazene reporter, DiaAlk, enables stable covalent modification of S-sulfinylated proteins from mammalian cells. Our chemoproteomic workflow permits site-specific quantification of dynamic fold-changes in sulfinylome modifications.
