Framework and components of a crystallization phase diagram. The ‘crystallization phase diagram’ is a useful operational framework for visualizing processes during crystallization. It is derived from the water-rich corner of an isothermal section of a pseudo-ternary phase body water–protein–reagent.
In this article I describe methods for measuring the phase diagram of an aqueous solution of a globular pro-tein, focusing on the crystalline (Section 2) and liquid phases (Section 3) which form. In Section 4 I describe how the phase diagram provides information which can be useful for protein crystallization.

A phase diagram provides information about crystal nucle-ation and growth conditions as a function of protein concentra-tion and thermodynamic parameters such as temperature, pH, and/or precipitant concentration. The portions of the phase diagram associated with crystallization can be divided into three regions.
The process of crystallization can be differentiated into two steps: (1) the nucleation process, and (2) crystal growth. With correctly selected conditions, crystal nucleation and growth can occur within the supersaturated regions of the phase diagram. (see Fig. 1).
